利用麸皮和木薯渣生产饲用木聚糖酶的黑曲霉固态发酵条件优化
Optimized solid-state fermentation of Aspergillus niger for production of xylanase using cassava dregs and bran
【摘要】 利用产木聚糖酶的黑曲霉(Aspergillus niger)GXH-1菌株对木薯渣和麸皮发酵,通过单因素和响应面试验优化产酶条件。最佳发酵条件为15.99%的木薯渣与84.01%的麸皮组成发酵基料;以1.60%的(NH4)2SO4为氮源;含水量为56.27%,KH2PO4和NaOH质量分数分别为0.12%和0.20%,接种量为4.0%;36.23℃发酵68h。在此条件下,木聚糖酶酶活力最高为1 258.74U/g,是优化前的3.26倍,优化效果显著。
【Abstract】 The cassava dregs and bran were fermented by Aspergillus niger GXH-1 strain to produce xylanase.Xylanase production conditions were optimized by single factor and response surface experiments.The optimum conditions for producing xylanase were as follows:the composition of culture medium was 15.99%cassava dregs and 84.01% bran,the nitrogen source was 1.60%(NH4)2SO4,the water content was 56.27%,the content of KH2 PO4 and NaOH were 0.12% and 0.20%,respectively,the inoculum was 4.0%,the fermentation temperature was 36.23℃ and the time was 68 h.Under the conditions,the enzyme activity of xylanase produced reach 1 258.74 U/g,which was 3.26 times higher than that before optimization.
